CBN is the primary oxidative degradation product of THC and is found in aged cannabis. Preclinical evidence from the SAMP8 mouse model shows CBN improves spatial learning and memory, with sex-specific metabolic benefits, by modulating mitochondrial dynamics and upregulating synaptic proteins. A zebrafish study demonstrated that CBN exposure during gastrulation causes dose-dependent developmental defects via CB2 receptor-mediated mechanisms, raising safety concerns for prenatal exposure. A human proof-of-concept trial (CUPID) is investigating 30mg and 300mg oral CBN for insomnia using polysomnography, but results have not yet been published. Evidence for CBN as a standalone sleep aid remains largely anecdotal and preclinical; robust human clinical data are lacking.
