§ 01 — EFFECTS
6 documented
Significant dose-dependent weight loss
Reduced HbA1c and improved glycemic control
Delayed gastric emptying
Reduced food intake and appetite suppression
Improved liver steatosis markers
Potential improvement in cardiovascular risk factors
§ 02 — RISKS
Dose-dependent increases in heart rate peaking at 24 weeks
Gastrointestinal adverse events including nausea, vomiting, and diarrhea
Two deaths reported in Phase 3 trial group (assessed as unrelated to study drug)
Study discontinuation due to adverse events in 2-5% of retatrutide recipients
Long-term safety profile not yet fully established
§ 03 — INTERACTIONS
Interaction data not yet available for this compound.
§ 04 — SCIENCE

Phase 2 and Phase 3 clinical trials demonstrate significant dose-dependent weight loss up to 24.2% at 48 weeks with 12mg and HbA1c reductions up to 2.02% at 24 weeks in adults with obesity and type 2 diabetes. Common adverse events are primarily gastrointestinal, dose-related, and mostly mild to moderate in severity. Dose-dependent increases in heart rate were observed, peaking at 24 weeks and declining thereafter. Long-term safety data from ongoing Phase 3 TRIUMPH trials is not yet available, and the quality of weight loss beyond BMI reduction requires further investigation.
