Coluracetam was originally developed by Mitsubishi Tanabe Pharma and later investigated by BrainCells Inc. in Phase IIa trials for anxiety and depression comorbid with MDD, showing some signal in a subset of patients. Preclinical studies suggest HACU enhancement improves cognitive function under choline-limited conditions. Human pharmacokinetic data are extremely limited, and no large-scale randomized controlled trials have been published. Its inclusion in reviews of glutamatergic antidepressants alongside ketamine reflects interest in its AMPA-mediated mechanisms, but clinical evidence remains preliminary and weak.
