4-AcO-DMT is a synthetic tryptamine that acts as a prodrug for psilocin, with in vitro metabolism studies identifying 15 metabolites and the hydrolysis product (psilocin) being most abundant. Animal drug discrimination studies show it fully substitutes for DOM, confirming hallucinogen-like effects, though 4-acetoxy compounds were less potent than 4-hydroxy analogues in vitro. A 2023 study found anti-inflammatory effects in human 3D EpiIntestinal tissue, reducing TNF-α and IFN-γ levels. Animal studies suggest its rewarding and withdrawal dynamics differ from classical drugs of abuse, potentially not following the opponent-process theory. However, human clinical data is extremely limited, and most evidence comes from animal models, in vitro studies, and survey data of recreational users.
